This study aims to analyze the characteristics of the three-tier diagnostic test instrument on cell biology material, describe the level of students' misconceptions, and evaluate students' responses to the implementation of the instrument. This research employs a descriptive quantitative approach with a case study method focused on 11th-grade science students at SMAN 10 Pekanbaru during the 2023/2024 academic year. Data collection techniques involved the use of a three-tier diagnostic test, observation, documentation, and validated questionnaires. The three-tier diagnostic test was used to identify students' misconceptions through three stages: objective questions, reasoning behind the answers, and students' confidence in their responses. The data obtained were statistically analyzed to categorize the levels of students' misconceptions into low, moderate, and high. The results of the study showed that 41.7% of students experienced low misconceptions, 56.6% had moderate misconceptions, and 1.7% had high misconceptions.
